What Line-Up does in Dragon Quest 3 Remake
The Line-Up menu can be found by opening the main menu and going to 'Miscellaneous', where the line-up is the fourth item from the top. Here you have the freedom to change your group to any order you want, although there are some jobs that work better in certain positions. Your composition determines the chance that a party member will be targeted in combatwith the most likely far left and the least likely far right.

While there's no wrong way to handle your party composition, the only effect is who gets targeted more often, like turn order is determined by the agility of each party member. When buying better armor or gear accessories, it's best to prioritize the man on the left, as he will take the most damage. The last spot in the party is also ideal for a healer as they usually have low defense but also need to stay awake to revive any downed party members.

How to change party leaders in Dragon Quest 3 Remake
Your party leader will be the character you control outside of combat and will control the other three party members. This is determined by whoever takes the leftmost spot in your lane, meaning that any party member can be your leader. Outside of the order your party members are in, there are no other changes with the party leader change, so it's best to focus on game effects and have your the tankiest character leading the group.
